The Position Genentech, Inc. seeks a Research Associate, Engineering Development (Cell Therapy) in its South San Francisco, California location. Duties: Serve as a researcher within the Individualized Medicines Team in Pharma Cell and Gene Therapy Group (“PTC”), which is responsible for the process development, manufacture and supply of personalized immunotherapies. Provide biological/chemical samples to Quality Control to support release and registration, transfer to manufacturing/production sites for use and storage and prepare for research and development activities. Evaluate biological/chemical equipment and the manufacturing environment in accordance with current Good Manufacturing Practices (cGMP) requirements and Roche corporate standards. Utilize sequencing platforms such as Oxford Nanopore for DNA assembly, cloning, screening, and sequencing. Perform and test process development experiments such as E.coli fermentation and lysate preparation to generate material for downstream plasmid purification. Research and analyze current scientific and industry trends in the field of gene therapy development. Implement new technology tools and instrumentation to optimize manufacturing and production operations. Support plasmid manufacturing for novel individualized cell therapies. Provide technical support to cell therapy process development teams in optimization of the cell therapy manufacturing process. Education and experience required: Bachelor’s degree in Biology or closely related field and 5 years of progressive, post-baccalaureate experience as Research Associate, Scientist, or related role. Special Requirements: Must have 5 years of experience with each of the following: Design and modify construct to optimize protein expression in different types of host cells including E. Coli, yeast, insect cell and mammalian cell; Express, extract and purify target proteins across different host cell types; Evaluate target protein expression level, purity, stability and aggregation by SDS-PAGE, FPLC, HPLC, Western Blot; Molecular Cloning of plasmids from primer design to PCR, Gibson assembly, transformation, colony screening to verification of sequence. Must have 3 years of experience with each of the following: Human primary T cell isolation and culture, lentiviral transduction, cell sorting and cytotoxicity assay (in-vitro killing assay); Flow cytometry for analysis of cell therapy product: panel design and flow analysis for testing activation state, phenotype of CAR T cell. May telecommute up to 2 days per week. Worksite: 1 DNA Way, South San Francisco, California, 94080. The expected salary range for this position based on the primary location for this position of California is $114,408 to $143,400 per year. Actual pay within the range will be determined based on experience, qualifications, geographic location, and other job-related factors permitted by law.
