We are seeking for a talented scientist with gene-to-structure experience to join our Structural Biology group within the Protein Science and Technology Department. The successful candidate will help accelerating Cryo-Electron Microscopy (Cryo-EM) and Crystallography workflows and will play a pivotal role supporting and implementing AI, protein engineering and epitope mapping approaches. This position requires excellent protein chemistry and Cryo-EM skills to individually contribute and guide protein expression, purification and structural characterization of recombinant proteins and complexes to support small molecule and biologics discovery. The Protein Science and Technology Department is responsible for all gene-to-structure work as well as hit-to-lead and lead optimization to study the mechanism of action of new and existing therapies.
The ideal candidate would have extensive experience working on a matrix team comprised of protein chemists, structural biologists, chemists, machine learning and data scientists to implement new structure and AI based technologies. The candidate is also expected to assist and oversee both scientific and operational aspects of our Cryo-EM and protein production laboratory to support a diverse portfolio of projects.
- Provide hands-on contributions to strengthen Incyte's internal capabilities in Cryo Electron Microscopy (Cryo-EM), Cryo Electron Tomography (CryoET), mechanism of action (MoA) studies, and structure-based drug discovery.
- Experience applying and/or implementing cryo-EM based methods for mapping of polyclonal antibody responses (Cryo-EMPEM) is highly desirable.
- Experience using cryo-EM data to identify and dock small molecule binding sites on targets is desirable.
- Provide hands-on contributions to design and purify stable protein samples for structural studies.
- Experience applying and/or implementing AI-based technologies to facilitate protein construct design and protein structure determination.
- Integrate and collaborate with different functions across the organization.
- Cryo-EM experience is required; crystallography experience is highly desirable but not required.
- Develop and implement Cryo-EM workflows.
Knowledge and Skills:
- Ability to apply AI-based techniques to facilitate protein construct design and protein structure prediction.
- Experience in designing and building mechanistic assays to define mechanism of action of various therapeutic approaches.
- Experience with multiparameter optimization of sample preparation.
- Experience working on a fast paced and dynamic environment to generate data that can impact program decisions.
- Established reputation with a track record in biochemistry, biophysics, and structural biology is highly desirable.
- Demonstrated independent thought/creativity in science with attention to details. Excellent communication (oral and written) in articulating goals, objectives, hypotheses, and results to the group and larger cross-functional project teams.
Qualifications (Minimal acceptable level of education, work experience, and competence)
- Ph.D. in structural biology, chemistry, biochemistry or related field with a minimum of 5 years of experience or BS/MS with 10 years of experience.
- Proven experience supporting drug discovery programs using structural biology and biochemistry.
- Good communication and interpersonal skills.
- Proven experience working with matrix teams
- Self-driven, creative and entrepreneurial with ability to support multiple programs simultaneously.
