About STAR0602 

STAR0602 is Marengo’s lead program, the first selective T cell activator generated from Marengo’s STAR platform; a library of antibodies targeting non-clonal variable (V)ß regions of the TCR fused to different co-stimulatory moieties. STAR0602 selectively targets a common Vb T cell subset present in all cancers and, by combining a novel non-clonal mode of TCR activation with a T cell co-stimulator in the same molecule, promotes expansion of a new population of clonally enriched, effector memory Vb T cells that turbo-charge immune system and promote a durable anti-tumor immune response. STAR0602 has undergone extensive preclinical testing, which demonstrates potent anti-tumor activity in both mouse and human ex vivo tumor models attributed to a distinct mechanism of action from existing cancer immunotherapies.

The Scientist II role within the “Antibody Technologies & CMC” function is a crucial position at Marengo Therapeutics, Inc. Reporting directly to the Head of our Protein Sciences group, the ideal candidate is a highly motivated, versatile, and experienced scientist in biopharmaceutical process research and analytical sciences. This role is instrumental in supporting both our discovery and development projects. 

As a Scientist II, you will play a pivotal role in our operations. Your responsibilities will include molecular biology, expression, and purification of various protein constructs for numerous projects across Marengo’s pipeline. You will also collaborate with CMC teams, providing valuable input on CMC projects. We expect you to have demonstrated hands-on experience in the production of various biologics, such as Fc-fusions, monoclonal antibodies, and bi and multi-specifics. A strong understanding of cutting-edge expression (Transient and Stable) and purification, as well as basic analytical processes, is also essential.

Requirements

Responsibilities:

· Develop methods to establish high-efficiency transient and stable cell production systems to support the production of reagents and therapeutic proteins. 

· Execute hands-on lab work to express, purify, and characterize novel protein therapeutics to support antibody screening and lead optimization in discovery programs

· Protein expression method development using shake flasks, 24 and 96 well plates. Support protein expression scale ranging from 1 mL to 20L.

· Lead process improvements for the lead candidate to support CMC work

· Develop expression and purification workflows to maximize productivity and efficiency

· Act as a Subject Matter Expert in Antibody production and process sciences and provide guidance to the research project teams.

· Analyze and present data, both in written reports and oral presentations.

· Manage collaborations with external CROs.

· Exhibit versatility in taking on new responsibilities within the Antibody Technologies group.

· Act as a champion and represent the Antibody technologies team in project team meetings. Participate in data presentations and cross-functional collaborations, with the opportunity to serve as ADPS lead.

Education and Experience:

· M.S. or PhD in Biochemistry, Molecular Biology, Cell Biology or a closely related field and 5  years experience in Molecular Biology, mammalian cell line development, antibody generation, and bioanalytical characterization is required

· Prior experience in pharma or biotech is required

· Experience in the production of complex Antibody-derived biologics using mammalian host systems and purification using a wide range of AKTA systems

· Strong background and hands-on experience in mammalian cell culture, stable cell line generation and process development

· Expertise and hands-on experience with protein purification methods used in discovery and process development. Expert knowledge of AKTA protein purification systems and chromatography resins

· Experience in characterization of biologics using advanced analytical techniques is a plus
