DescriptionIncumbent will perform biochemical and genetic studies of novel filaments in the budding yeast, S. cerevisiae. Responsibilities include a wide variety of standard repetitive procedures such as generating particular yeast via genetic techniques and analyzing the progeny by a combination of live imaging, immunoblotting, enzymatic assays, immunofluorescence and other techniques.
The incumbent will also be responsible for generating recombinant DNA constructs that the individual and other members of the laboratory will use for protein expression as well as for making yeast strains. The incumbent will also be responsible for the expression and purification of filament forming proteins and the development of assays for polymerization.
The incumbent will also be responsible for the maintenance and feeding of the yeast stocks for the laboratory. Laboratory management duties include ordering supplies, lab organization, maintenance of computer and laboratory equipment, and providing work direction to undergraduate students.
Qualifications
  • Theoretical and practical knowledge of biochemistry, molecular biology, immunology and genetics, including relevant laboratory techniques such as genomic technologies using RNAi and microarray analyses, acquired through a Bachelor's degree in Biology or a related field or experience, and/or equivalent combination of education and laboratory experience.
  • Theoretical knowledge and previous experience with a variety of small- and large-scale molecular biology techniques, including: DNA preparation, PCR, in vitro transcription, nucleic acid purification, enzyme restriction digests, gel electrophoresis, ligation, transformation, screening for clones, plasmid construction, isolation and analysis, preparation of radioactive probe for hybridization and autoradiagraphy, sequencing, microarray hybridizations, protein purification, immunofluorescence, and microinjection.
  • Proven ability to express and purify proteins using affinity and conventional chromatography
  • Proven ability to develop enzyme assays for protein activity including assays for the ability to self-assemble.
  • Proven ability to execute multi-step assays and multi-generational yeast crosses.
  • Working knowledge to operate various lab instruments including: bright-field and fluorescent microscopes, centrifuges, spectrophotometer, hemocytometer, micropipetting equipment, computer for maintaining stock inventories and protocols.
  • Ability to work independently with supervision in efficient and organized manner.
  • Familiar with PC computers and software such as Microsoft Office, with skill in use of PC computer databases for quality control, tracking, and data input. Willingness and ability to learn new hardware and software as needed.
  • Skill, ability, and willingness to learn new laboratory techniques, lab equipment, and computer hardware/software rapidly and as needed. Ability and willingness to read scientific papers as directed in order to stay abreast of developments within the field and relating to current experiments.
  • Knowledge and ability to order lab supplies and equipment, and to proactively and appropriate facilitate the maintenance of all lab supplies and equipment.
  • Excellent oral and written communication skills with the ability to maintain clear, concise lab notes and to communicate experimental results effectively on the telephone or via email.
  • Positive and motivated attitude, and ability to work congenially and collaboratively with others in growing lab environment.
  • Knowledge of MSDS's (Material Safety Data Sheets), and the hazard communication and Injury and Illness Prevention Program. Familiarity with a Chemical Hygiene Plan addressing safety precautions including appropriate clothing and shoes, safe handling of chemicals, use of protective items such as gloves and goggles. Willingness and ability to keep safety training current and consistent with research involvement.
  • Demonstrated knowledge of cell and protein analyses, including experience with Western blot analysis and subcellular localization by fluorescent microscopy.
  • Previous experience with S. cerevisiae yeast and familiarity with genetic crosses.
  • Skill, knowledge and ability to provide the full range of supervisorial duties (select, train, work-direction, and disciplinary action up to and including release) for undergraduate student lab employees.
  • Willingness and demonstrated ability to embrace and consistently uphold the principles of community for a collaborative work environment.
SPECIAL CONDITIONS
  • Must be willing to work with radioisotopes.
  • Willingness to be flexible with work hours to include occasional evening or weekend work as required by particular experiments.
  • Background check required.
Pay Transparency ActAnnual Full Pay Range: $53,766 - $64,143 (will be prorated if the appointment percentage is less than 100%)
Hourly Equivalent: $25.75 - $30.72
Factors in determining the appropriate compensation for a role include experience, skills, knowledge, abilities, education, licensure and certifications, and other business and organizational needs. The Hiring Pay Scale referenced in the job posting is the budgeted salary or hourly range that the University reasonably expects to pay for this position. The Annual Full Pay Range may be broader than what the University anticipates to pay for this position, based on internal equity, budget, and collective bargaining agreements (when applicable).
